In this review, we provide an update on the dynamic distribution of neutrophils across tissues in health and disease, and emphasize differences between humans and model organisms. We further highlight issues to be addressed to exploit the unique features of neutrophils in the clinic.

Life cycle of a neutrophil cell. Approximately 10 11 PMNs are generated in the bone marrow via granulopoiesis every day. Attracted by cytokines, PMNs are consecutively released into the blood stream and thus into systemic circulation.

Recent advances in neutrophil biology have clarified the life cycle of these cells and revealed striking differences between neonatal and adult neutrophils based on fetal maturation and environmental factors.

In this review, we highlight early and recent work that will help the reader to appreciate our current view of the neutrophil life cycle -from maturation to elimination-, and how neutrophils behave and dynamically modulate tissue immunity, both in steady-state and in disease.

To achieve this, it is mandatory to understand the basic rules guiding the life cycle of neutrophils in health and disease (Box 1). This review focuses on the recent new data in this area.

First a general description of the exciting life of a neutrophil from birth to death is given. Different parts of the neutrophil life cycle are discussed as well as the kinetics. Several functions of neutrophils and the consequences of these functions to their life span will be discussed.
